A method note deals with a specific point of execution. It does not present a discipline but a decision: how to design a panel, how to prepare a sample, how to read an evaluation result. It is written for whoever is going to do the work, or has to judge what has been done.
Each closes on a checklist, and on what cannot be recovered once the experiment has been run.

The notes
- Designing a cytometry panel: spectral vs conventional — a panel is not a list of antibodies but an optical trade-off, and spectral unmixing has a pitfall of its own.
- Sample preparation for scRNA-seq — dissociation alters composition and manufactures a signal; what is lost at that step is not recovered at analysis.
- Evaluating a candidate preclinically: effect, exposure, mechanism — why an efficacy result alone establishes nothing, and why a negative result without exposure data is uninterpretable.
