Das In-vivo-Experiment bleibt ein entscheidender Schritt der translationalen Forschung: Auf Ebene des gesamten Organismus werden die Wirksamkeit, die Sicherheit und die physiologische Relevanz einer therapeutischen Hypothese überprüft. Es bildet das Gegenstück, auf Ebene des Organismus, zu der in der Zellbiologie geleisteten Arbeit.
Inovarion setzt ein breites Spektrum präklinischer Modelle ein. Die genetisch veränderten Mausmodelle – konditionale und gewebespezifische Knock-outs, Knock-ins, transgene, Doppel-Knock-outs – bilden deren Kern. Das Labor greift namentlich auf humanisierte Modelle zurück, die menschliche Proteine tragen: So hat es zu einem vollständig humanisierten Mausmodell der Von-Willebrand-Erkrankung beigetragen, einer wertvollen Plattform, um neue therapeutische Optionen zu bewerten[3]. Hinzu kommen dedizierte Krankheitsmodelle (mdx und Doppel-Knock-out für die Duchenne-Dystrophie, Hämophilie- und Blutungsmodelle, durch fettreiche Ernährung induzierte Adipositas, Lithium-Pilocarpin-Epilepsie[1]), patientenabgeleitete Xenotransplantate und orthotope Modelle in der Onkologie, der Zebrafisch für die In-vivo-Bildgebung und die Optogenetik sowie der nichtmenschliche Primat für die Neurowissenschaften.
Diese Modelle dienen der In-vivo-Validierung von Zielstrukturen und Therapien in allen Bereichen: Onkologie (Xenotransplantate, orthotope Modelle, maligne Transformation beim Zebrafisch[7]), Hämatologie (Hämophiliemodelle, humanisiertes von-Willebrand-Modell), seltene Erkrankungen (Duchenne-Dystrophie), Stoffwechsel (adipozytenspezifische Knock-outs), Kardiologie (BMP9[11]/BMP10-Modelle) und Neurologie (Epileptogenese, nichtmenschlicher Primat). Ihr Wert liegt in ihrem Realismus: humanisierte und konditionale gewebespezifische Modelle, oft gekoppelt an iPSC und Transkriptomik, um den Phänotyp des Organismus mit den molekularen Mechanismen zu verknüpfen.
Inovarion konzipiert und nutzt diese Modelle in Kohärenz mit der vorgelagert geleisteten In-vitro- und bioinformatischen Arbeit – die Erzeugung der genetischen Modelle fällt der Molekularbiologie zu, ihre Auslesung der Bildgebung und der Bioinformatik. Diese Kontinuität, vom Modell bis zu seiner Auslesung, dient den Projekten der Partner, vom In-vivo-Proof-of-Concept bis zur Bewertung von Wirkstoffkandidaten.
